Adeno-associated virus (AAV) is a small, non-pathogenic human DNA virus. AAV-based viral vectors are widely used in the field of gene therapy. Viral vectors encapsulate, transport and deliver therapeutic genes into cells. These genes can replace or repair faulty genes, providing potential cures for various genetic disorders and diseases.

Detection and characterisation of AAVs is essential throughout manufacturing of these advanced therapy medicinal products, to ensure their critical quality attributes meet stringent regulatory requirements for efficacy and safety.  A particular challenge identified by manufacturers is in determining the levels and quality of the AAVs at various stages in the cell culture phase. Of critical importance is the need for rapid analysis of viral load during culture optimisation and subsequent virus purification.

The technical note outlines the methodology with the selectivity and sensitivity required to meet the need for rapid enumeration of AAV titres.
